KUMC ADC blood draw protocol

Draw occurs at initial visit and at T5

REG Draw and MITO Draw combined into one draw
11132 REG/MITO Draw (2 red tops, 7 yellow tops)
•  Serum 6 cryovials (0.5 ml/tube)
•  Platelet-Rich plasma 6 cryovials(0.5 ml/tube)
•  Platelet-free plasma 6 cryovials(1.2 ml/tube)
•  Buffy coat 10 cryovials(0.5ml/tube)
•  Whole blood 2 cryovials (0.5 ml/tube)

Plasma was spun at 200xG for 10min and 5ml are removed and then spun again 
at 16,000XG for 10min. The remaining Plasma was aliqouted into vials at 1.2ml.  
The plasma that was spun twice was aliqouted into 0.6ml aliquots.

   
3/30/15—New processing protocol started (initial visit and as needed)
   
7/28/15—11132 protocol approved to obtain blood samples on initial visit and 
    T5 participants (according to the processing procedure adopted 3/30/15).

9/14/15—Zaid adjusted the platelet processing technique to ensure more platelets 
    were captured in the sample instead of other cells.

REG Draw:
11132 RGE Draw (2 red tops, 2 yellow tops, 2 purple tops)
•  Serum 12 cryovials (0.5 ml/tube)
•  Plasma 6 cryovials(1.2 ml/tube)
•  Buffy coat 3 cryovials(~1 ml/tube)
•  Buffy coat MGM Core DNA cryovials (4 aliquots, 500ul each )
•  MGM Core Whole blood 2 cryovials (0.5 ml/tube)

REG:  Plasma was spun at 1000xG for 12min and then aliquoted into vials at 1.2ml

 
11132 MITO Draw (5 yellow tops) 
•  Platelet-Rich plasma 6 cryovials(0.5 ml/tube)
•  Platelet-free plasma 6 cryovials(1.2 ml/tube)
•  Buffy coat 10 cryovials(~0.5 ml/tube)
•  Whole blood 10 cryovials (1 ml/tube) If subject agreed for the whole blood portion. 

MITO:  Plasma was spun at 200xG for 10min and then spun again at 16,000XG for 10min. 
   After that it was aliqouted into vials at 1.2ml.
